By disrupting mitochondrial energetics, metformin alters passive transport through the nuclear pore complex. This prevents RagC from transiting through the nucleus, and acquiring the GDP-bound active state necessary for mTORC1 activation. Inhibition of mTORC1 increases transcription of ACAD10, in part through SKN-1/Nrf2. ACAD10 inhibits growth and extends lifespan in C. elegans, and reduces human cancer cell viability.
