Gugglesterone prevents inflammatory changes in HNPCECs stimulated with LPS. (A) Growth-arrested HNPCECs pretreated with or without gugglesterone (40 μM) were incubated with 1 μg/mL LPS for 24 hours. Expression of Cox-2, iNOS, AR and phospho-p65 from cell lysates was determined by Western blot analysis using specific antibodies. (B) Growth-arrested HNPCECs pretreated with or without gugglesterone (40 μM) were incubated with 1 μg/mL LPS for 0 to 120 minutes. PI3K and Akt from total cell lysates were determined by Western blot analysis using respective phosphospecific antibodies. (C) Levels of TNF-α in the culture media were determined with ELISA kit.
