MOSCOW (RUSSIA) WOMEN WITH POSITIVE SKIN PRICK-TESTS: COLOSTRUM AND BREAST MILK CYTOKINES AS AN ADOPTIVE PROTECTIVE MECHANISM FOR THE OFFSPRING Imperial College London Pirogov Russian National Research Medical University Marina Treneva1, Daniel Munblit 2, Alexander Pampura1 1 Research and Clinical Institute for Pediatrics at the Pirogov Russian National Research Medical University, Moscow, Russia  2 Imperial College London, London, United Kingdom Research and Clinical Institute for Pediatrics (Moscow) Methods: 277 Moscow women were recruited as a part of an International (Great Britain, Russia, Italy) study assessing immune composition of colostrum and breast milk. 19,4% of Moscow women were sensitized to one or more allergens (D.pteronissinus, Cat, Birch pollen, Grass pollen, Ryegrass, Hazelnut, Peanut, Egg, Cow milk). Electrochemiluminiscence of colostrum (204 samples) and breast milk (153 samples) for HGF, IFN-γ, IL-2, IL-4, IL-5, IL-10, IL-12, IL-13, TGF-β-1, TGF-β-2, TGF-β-3 was performed in a Flow Cytometry Lab, Imperial College Background: There is some evidence to suggest that colostrums/breast milk immunological composition have potential in infant gut immunity modulation early in life Results: According to univariate analysis colostrum and/or breast milk levels of TGF-β-1, TGF-β-3, HGF, IL-5, IL-10 and IFN-γ were higher in women with positive skin prick-tests. No difference in IL-2, IL-4, IL-12, IL-13, TGF-β-2 levels was revealed in relation to positive skin prick tests. Table 1 contains results of all examples analysis without adjustment. Table 1 Reliable distinctions in colostrum and breast milk cytokine composition in Russian women depending on skin prick-test papule diameter Objective:  to assess levels of cytokines in colostrum and breast milk in women with sensitization. Absolute values Lg of absolute values colostrum breast milk Papule ≥ 1 mm to one or more allergens no difference in HGF, IFN-γ, IL-2, IL-4, IL-5, IL-10, IL-12, IL-13, TGF-β-2 no difference in IL-2, IL-4, IL-10, IL-12, IL-13, TGF-β-1, TGF-β-2, TGF-β-3; reliable distinctions in HGF p=0,037; IFN-γ p=0,042 no difference in HGF, IFN-γ, IL-2, IL-4, IL-5, IL-10, IL-12, IL-13, TGF-β-2; reliable distinctions in TGF-β-1 р=0,029 no difference in IL-4, IL-5, IL-10, IL-12, IL-13, reliable distinctions in HGF p=0,020; IFN-γ p=0,002 Papule ≥ 3 mm to one or more allergens no difference in HGF, IFN-γ, IL-2, IL-4, IL-5, IL-10, IL-12, IL-13, TGF-β-2 no difference in IFN-γ, IL-2, IL-4, IL-10, IL-12, IL-13, TGF-β-1, TGF-β-2, TGF-β-3; HGF p=0,013 no difference in HGF, IFN-γ, IL-2, IL-5, IL-10, IL-12, IL-13, TGF-β-2; reliable distinctions in TGF-β-1 р=0,003; TGF-β-3 р=0,040 no difference in IL-4, IL-5, IL-10, IL-12, IL-13, TGF-β-1, TGF-β-2, TGF-β-3; reliable distinctions in HGF p=0,012; IFN-γ p=0,005 Conclusion: colostrum and breast milk cytokine and growth factors in Moscow women allow to assume the lack of IL-4 and IL-13 shift in relation to sensitization. There may be an assumption that the increased levels of growth factors, IL-5, IL-10 and IFN-γ revealed could be the adoptive mechanisms in sensitized women to protect their offspring from an allergy, according to the unique regenerative nature of a human body. This hypothesis should be tested further to exclude influence of maternal and environment factors on results of the analysis, in connection with heterogeneity of the analyzed groups.
