Structural features of the p53 gene and its encoded protein. The gene encodes a 53-kDa nuclear phosphoprotein. The 393 amino acids are spread over 11 exons. The transcription activation site (TAS), heat shock protein binding site (HSP), SV40 large T antigen binding sites (SV40), nuclear localization signal (NLS), oligomerization domain (OLIGO), and phosphorylation sites (P) all identify potential functional regions. The five evolutionarily conserved domains (HCD I-V) correspond closely to the regions most frequently mutated in sporadic cancer [hotspot regions (HSR A-D)]. Sites of binding of the extracellular E1B 55-kDa protein of adenovirus type 5 (E1B) and the E6 gene product of human papillomavirus types 16 and 18 (E6) are also indicated, as is the position of the MDM2 gene product-binding site (MDM2).
