Dissecting Microscopes
Introductory Lesson
2
Always carry a microscope with two hands: one on the arm and one at the base for support
One partner (or period) will remove from storage shelf and set up. One partner(or period) will break down, cover and wrap cords & return to storage shelf. ****unplug microscope by grasping the plug not the cord!!!****
3
Parts of a Dissecting Microscope
This is the ocular lens (x10 magnification) Total magnification is always ocular x objective This is the objective lens: (x1, x2 and x3 magnification) Use light from above, below or both together Put your perti dish here on the stage
4
Microscope Protocol Always begin, end and store a microscope with the lowest power objective in place for dissecting microscope lower the head all the way between uses Always begin, end and store a clean microscope Clean all lenses and the stage with lens paper only Always begin, end and store a covered microscope with a wrapped cord. Always unplug a microscope by grasping the plug not the cord!!!

Dissecting Microscopes
Stereo microscopes have two separate light paths transmitting the image under study to each eyepiece. This takes advantage of our binocular vision Allows you to perceive object in three-dimensions Gives you a feeling of depth to the object
6
How to use the dissecting microscope
7
How to use the dissecting microscope
Adjust for your own interocular distance by spreading apart or squeezing together the oculars (as you would with binoculars) until a single three-dimensional image is seen. Use the focus knob on the side to raise the microscope body until the specimen image is the sharpest.

How to sketch what you see in the microscope
Anyone can learn to sketch a microscope slide! You only need white paper, pencils, erasers and rulers You DO NOT need to have artistic talent to create a decent sketch. This is a myth. Relax and have confidence that you can create a representational sketch. An eraser it all it takes to make improvements to your drawing. Sketches come to life when you add highlights, shadows and color. For a pencil sketch, separate areas into white, light, medium and dark grey and black.
9
How to sketch what you see in the microscope
Requires an open-mind, patience, attention to detail and a willingness to use basic drawing principles like perspective, size, shape and negative space. Sketching will provide you with a better understanding, as you study the details of the image you see through the lens these details that may be overlooked in a photo.
10
How to sketch what you see in the microscope
Use a petri dish to draw a circle on your paper. Note the specimen, date, mounting technique, magnification and other related information. Establishing a habit of labeling your drawings while discovering how to sketch a microscope slide will enable you to keep your drawing organized.
11
How to label your drawing
Put a title here & underline it Make a circle with a petri dish to represent your field of view Make horizontal lines with a ruler to the right for your labels Write your total magnification here

Today Find two radish seeds at early and later stages of germination.
View with microscope and submit sketches. Label the following: Primary root Root hairs Leaves (or shoots) Seed (what’s left of seed coat)
