Microscope Review This presentation reviews content we have already completed in class, in a variety of assignments. This is NOT new. Practice and review and ASK QUESTIONS

2 Introduction to the Microscope
Safety and Care Parts Focusing Students will have their microscopes out, for reference as we go through each of the steps. As an introduction, students will be asked what kinds of things they can do with this tool.

3 Safety and Care Store in a cool, dry place Have cord folded and banded
Cover with dust cover Stage all the way down Low power objective at stage Eyepiece over arm Do not stack

4 Microscope Care Always carry with 2 hands
Only use lens paper for cleaning Do not force knobs Always store covered Keep objects clear of desk and cords Teacher demonstrates how to hold the microscope, where the lens paper is located and how to use it. Students will be invited to turn the knobs and observe the stage as it moves up and down. Teacher will demonstrate how to store the microscope.

7 Using the Microscope Place the Slide on the Microscope Stage
Use Stage Clips Click Nosepiece to the lowest (shortest) Objective Move stage closest to objective Look into the Eyepiece Use the Coarse Focus THEN, Use the fine focus Use the pointer to keep track of your specimen Give students a slide from the “common things” set, each student will practice focusing and changing objectives.

8 Using High Power Follow steps to focus using low power
Click the nosepiece to the longest objective Do NOT use the Coarse Focusing Knob Use the Fine Focus Knob to bring the slide Have students exchange slides so they can look at different things, walk them through using the high power objective to focus slides. Emphasize not using the coarse objective during this process, as it will crack the slides. What can you find on your slide?

9 Microscope Investigation
You have observed plant, animal, protists. You have sketched and photographed. Can you see any organelles? If so, which ones? Why or why not?

10 Questions 1. State 2 procedures which should be used to properly handle a light microscope. 2.      Explain why the light microscope is also called the compound microscope. 3.      List the procedures for preparing a wet mount slide.

13 Protists, protozoa Protozoa are single-celled eukaryotes (organisms whose cells have nuclei) that commonly show characteristics usually associated with animals, most notably mobility and heterotrophy. They are often grouped in the kingdom Protista (protists) together with the plant-like algae and fungus-like water molds and slime molds.

A prokaryotic cell has five essential structural components: a nucleoid (region) (DNA),ribosomes,  cell membrane,  cell wall, and some sort of surface layer, which may or may not be part of the wall.

There are three regions: appendages  (attachments to the cell surface) in the form of flagella and pili (or fimbriae); a cell envelope consisting of a capsule, cell wall and plasma membrane; and a cytoplasmic region that contains the cell chromosome (DNA) and ribosomes  (Figure 1).  Figure 1
