Activation of the IL‑15 pathway via JAK‑1, JAK‑3 and SYK phosphorylation protects against the development of severe glomerulonephritis. This protective effect depends on the expression of γc (which is absent in Rag2−/−Il2rg−/− mice) and other subunits of the IL‑15 receptor (such as the IL‑2 receptor subunit β, IL‑2‑Rβ, which is absent in Rag2−/−Il2rg−/− mice) in intrinsic renal cells but not in immune cells. Natural killer (NK) cells, which are known to be protective in glomerulonephritis and are absent in Rag2−/− mice, do not seem to have an important role in this process but might contribute to protection from tubulointerstitial lesions.
