(B) Periodic acid Schiff (PAS) staining. Mice were euthanized, and the thyroids dissected, fixed in buffered formalin, and embedded in paraffin. Thyroid sections (5 μm) were mounted on glass slides, de-paraffinated, and hydrated. For histological analysis, sections were stained with H&E, following a standard protocol. Glycoproteins were detected using PAS staining. Sections were stained with 0.5% periodic acid for 30 minutes and with Schiff's reagent for 20 minutes and then rinsed in running tap water for 5 minutes. Nuclei were counterstained with hematoxylin for 3 minutes. Sections were rinsed in running tap water, dehydrated, cleared, and mounted. Reproduced with permission from Senou et al. (20).
