Fig. 1 Time courses of (A) Gly-Sar and (B) glycine uptake into BBMVs from control and 5-FU–treated rats. (A) BBMVs from control (○) and 5-FU–treated (●) rats were preloaded with 100 mmol/L K2SO4 and 50 mmol/L HEPES-Tris (pH 8.5) and then incubated for the indicated times at 20°C in the presence of 300 mmol/L mannitol, 50 mmol/L HEPES-Tris/2-(N-morpholino)ethanesulfonic acid (pH 5.5), and 1 mmol/L [3H]Gly-Sar. (B) Glycine transport in BBMVs from control (○)and 5-FU–treated (●) rats was measured in the presence of an Na+ gradient as previously described.12 Data in A and B are means ± SE of 6 rats. *P < 0.01 vs. control rats. Fig. 2 Lineweaver–Burk plot of Gly-Sar transport by ileal BBMVs isolated from (A) control and (B) 5-FU–treated rats. The condition of transport assay was shown in Figure 1. Data were obtained with a 1-minute incubation period. V, nmol Gly-Sar · min−1 · mg protein−1; S, millimolar concentration of Gly-Sar. Values represent means of 8 determinations. Fig. 3 Immunoblot analysis of PepT1 in rat small intestine. Male Wistar rats with body weight of 220 g were maintained on a normal diet (Purina Chow) for 7 days. After they were deprived of food for 24 hours, the rats were killed and the small intestine was removed. (A) PepT1 immunoreactivity in BBMVs from duodenum (lane 1), jejunum (lane 2), and ileum (lane 3) of control rats. Each lane contains 50 μg of protein. (B) Parallel staining was carried out with the antibody mixed with the antigen peptide (50 μg/mL). Lane 4, preabsorbed PepT1 antibody; lane 5, PepT1-specific antibody. Fig. 4 Effects of 5-FU treatment on PepT1, sucrase, and SGLT1 expression in ileal BBMVs prepared from control and 5-FU–treated rats. BBMVs were isolated from rats 3 days after administration of 5-FU and then subjected to immunoblot analysis (30 μg protein/lane) with antibodies to (A) PepT1, (B) SGLT1, or (C) sucrase. Lane 1, control; lane 2, 5-FU treated. Gastroenterology , DOI: ( /S (98) ) Copyright © 1998 American Gastroenterological Association Terms and Conditions

Fig. 5 Immunostaining of the ileum of normal rats with antibodies to PepT1. (A) PepT1 immunoreactivity can be seen only in the BBM of mucosal epithelial cells. (B) No immunostaining was detected with the preabsorbed PepT1-specific antibodies with the antigenic peptide (bars = 20 μm). Fig. 6 Immunostaining of the ileum of control and 5-FU–treated rats with antibodies to (A and C) sucrase or (B and D) PepT1. (A) Sucrase immunoreactivity is detected in the BBM of mucosal epithelial cells in the tip and middle regions of villi in control rats. (B) Three days after administration of 5-FU, sucrase immunoreactivity largely disappeared from villi. (C) PepT1 immunoreactivity in control rats. (D) PepT1 immunoreactivity was still prominent in all mucosal epithelial cells of villi 3 days after treatment of rats with 5-FU (bars = 20 μm). Fig. 7 Effects of 5-FU on expression of nutrient transporters in rat ileum. Northern blot analysis was performed on total RNA (20 μg) from the intestine of 5-FU–treated animals. (A) PepT1, (B) dipeptidyl peptidase IV, (C) sucrase-isomaltase, and (D) SGLT1. Cont, control. Fig. 8 Relative levels of intestinal nutrient transporter mRNAs. The relative concentrations of PepT1, SGLT1, neutral basic amino acid transporter (NBAT), 4F2hc, high-affinity glutamate transporter (EAAC1), Na+-dependent phosphate transporter (Glvr-1), sucrase-isomaltase (S-I), and dipeptidyl peptidase IV (DPP IV) were determined by densitometric scanning of autoradiographs. Results are given as means ± SE of the relative percentage of control. (A) Jejunum; (B) ileum.
