2 Fig. 1 Secretory phospholipase A2 steady state mRNA levels in mouse lungs and human cell lines. (A) CFTR−/− mice were sensitized/challenged with PBS or Af. Real-time RT-PCR was performed from whole lung mRNA for groups II, V, X and XII PLA2s: Pla2g2c, Pla2g2d, Pla2g2e, Pla2g5, Pla2g10 and Pla2g12b. Data points are the means of 2−ΔΔCT±SEM (7≤n≤10). * indicates p≤0.05 as compared to PBS-sensitized mice. (B) Real-time RT-PCR of PLA2G2D following a 12h treatment of S9 and IB3.1 cells with the indicated inflammatory stimuli. Data points are the means of 2−ΔΔCT±SEM (n=3). * and + indicate p≤0.05 as compared to IB3.1 control and treated S9 cells, respectively. 3 Fig. 2 Group IV cytosolic phospholipase A2s steady state mRNA levels in mouse lungs. CFTR−/− mice were sensitized/challenged with PBS or Af as described in Fig. 1. Real-time RT-PCR was then used to determine the steady state mRNA levels from whole lungs for the group IV PLA2s, Pla2g4a, Pla2g4b and Pla2g4c. * indicates p≤0.05 as compared to PBS-sensitized mice. 4 Fig. 3 Gene expression analysis of enzymes involved in prostanoid synthesis in mouse lungs. CFTR−/− mice were sensitized/challenged as described in Fig. 1, and real-time RT-PCR on whole lung mRNA was performed for Ptgs1, Ptgs2, Ptges, Tbxas1, Ptgds, and Ptgis. Data points are the means of 2−ΔΔCT±SEM (7≤n≤10). Journal of Cystic Fibrosis  , DOI: ( /j.jcf ) Copyright © 2012 European Cystic Fibrosis Society. Terms and Conditions

5 Fig. 4 Gene expression analysis of enzymes in the lipoxygenase pathway in mouse lungs and human cell lines. (A) CFTR−/− mice were sensitized/challenged with PBS or Af, and whole lung mRNA was analyzed by real-time RT-PCR for lipoxygenase gene expression (Aloxe3, Alox5, Alox5ap, Alox12, Alox12b, Alox15, Alox8, and Alox12e). Data points are the means of 2−ΔΔCT±SEM (7≤n≤10). * indicates p≤0.05 as compared to PBS-sensitized mice. (B) Steady state mRNA levels of human ALOX15 in response to the indicated stimuli in S9 or IB3.1 cells. Data points are the means of 2−ΔΔCT SEM (n=3). (C) Steady state mRNA levels of human ALOX15B in response to the indicated stimuli in S9 or IB3.1 cells. Data points are the means of 2−ΔΔCT±SEM (n=3). * and + indicate p≤0.05 as compared to control and S9 cells, respectively. 6 Supp. Fig. 1 Immune response in a model of Af sensitization/challenge in inbred mouse strains. C57BL/6 mice were sensitized with PBS, ovalbumin (OVA) or Af extract on days 0 and 14 and subsequently challenged on days 28–30 with Af in both PBS and Af-sensitized animals or OVA. Gene specific real-time RT-PCR analysis was performed on RNA extracted from whole lungs. Graphs represent steady state mRNA levels of (A) IL-4 and IL-13 (B) IL-1β, IL-2, IL-3, IL-6 and IL-10 and (C) GM-CSF, TGF-β, eotaxin-1 and KC gene expression. Cyclophilin A used as an internal control. Data points are the means of 2−ΔΔCT±SEM (n≥3). * and + indicate p≤0.05 as compared to PBS- and OVA-sensitized/challenged mice, respectively. 7 Supp. Fig. 2 Diagram of eicosanoid metabolism. The enzyme families within the arachidonic acid pathway are shown in bold with the corresponding specific enzymes (which were evaluated in this study by real-time RT-PCR) in the shaded gray boxes with the products produced from these reactions shown in the black boxes. Human gene names are used in this figure, while the corresponding mouse gene names are provided in Supplemental Table 2. AA is liberated from the phospholipid membrane through the action of the phospholipase A2 enzymes (both cytosolic and secretory). AA is then utilized by either the cyclooxygenase or lipoxygenase pathway enzymes. The cyclooxygenases produce PGH2 which is converted by specific synthases to prostaglandins or thromboxane A2; the lipoxygenases produce H (p)ETEs, HETEs, cystLTs, LTA4, LTB4, and lipoxins. 8 Supp. Fig. 3 Comparative response of group IV phospholipase A2 steady state mRNA levels to OVA and Af in C57BL/6 mouse lungs. Mice were sensitized/challenged with PBS, OVA or Af as described in Supp. Fig. 1. Real-time RT-PCR was performed from whole lung mRNA for group IV PLA2s: Pla2g4a, Pla2g4b, and Pla2g4c. Data points are the means of 2−ΔΔCT±SEM (n=5). * indicates p≤0.05 as compared to PBS-sensitized mice. Journal of Cystic Fibrosis  , DOI: ( /j.jcf ) Copyright © 2012 European Cystic Fibrosis Society.
