1 Proximal signaling events in FcɛRI-mediated mast cell activation

2 Fig 1 FcɛRI-mediated activation of mast cells. Interaction of FcɛRI-bound IgE with a multivalent antigen crosslinks FcɛRI on the cell surface leading to Syk activation. Signals from Syk are rapidly transduced, resulting in degranulation, leukotriene production, and cytokine/chemokine gene transcription. The release of these inflammatory mediators contributes to the acute and chronic symptoms associated with allergic diseases. PLA, Phospholipase A. 3 Fig 2 The SLP-76/LAT multimolecular complex in mast cell activation downstream of FcɛRI. The SLP-76/Gads/LAT interaction forms a multimolecular complex that localizes key signaling molecules in a spatial and temporal manner. Collectively, these events cooperate to induce degranulation and cytokine/chemokine and leukotriene release. The phosphatases SHP-1 and SHIP negatively regulate mast cell activation. Solid and dashed arrows represent positive and negative regulation of the downstream target, respectively. 4 Fig 3 Mast cell activation by SLP-76/LAT-independent pathways. Two src PTKs (Lyn and Fyn) phosphorylate Syk. In the presence of Lyn, the role of Fyn is less pronounced because of negative regulation by Lyn. Activation of the PI3K/Akt and ERK pathways occurs independently of SLP-76/LAT. Solid and dashed arrows represent positive and negative regulation of the downstream target, respectively.
