2. Overnight Culture Suspension Step 1
Label a sterile 50 ml Conical tube with your name and date.

3. Overnight Culture Suspension Step 2
Do a sterile transfer of one sterile pipette full of LB broth
containing ampicillin antibiotic into the Conical tube.

4. Overnight Culture Suspension Step 3
Locate a well-formed colony on your plate of transformed bacteria.
Use a sterile inoculating loop, bent paper clip, or toothpick to scrape
up the colony and transfer it to the LB broth in the tube.

5. Overnight Culture Suspension Step 4
Incubate the tube for 24 hours in a 37oC shaking water bath or
48 hours in a 37oC incubator with occasional shaking by hand
(shake the tube a couple of times an hour when possible).

6. Overnight Culture Suspension Step 5
After the overnight growth period is complete, proceed to part II,
Plasmid Purification.
