Novel Prognostic Markers in the Serum of Patients With Castration-Resistant Prostate Cancer Derived From Quantitative Analysis of the Pten Conditional Knockout Mouse Proteome  Martin Kälin, Igor Cima, Ralph Schiess, Niklaus Fankhauser, Tom Powles, Peter Wild, Arnoud Templeton, Thomas Cerny, Ruedi Aebersold, Wilhelm Krek, Silke Gillessen  European Urology  Volume 60, Issue 6, Pages (December 2011) DOI: /j.eururo Copyright © 2011 European Association of Urology Terms and Conditions

2 Fig. 1 Analysis of 79 proteins in sera of castration-resistant prostate cancer (CRPC) patients reveal 11 novel candidate biomarkers for survival. (A–O) We applied glycocapture to 57 CRPC patients to analyze 76 peptides representing 66 different proteins by SRM. Moreover, we established 13 ELISAs and collected from the records 8 further variables for a total of 87 variables quantified in a total of 57 patients. We subjected the resulting data matrix to survival analysis and found 15 variables to be significantly correlating with survival. Threshold protein concentration (tr) that separates prognostic groups is indicated. PVRL1=poliovirus receptor-related 1; PRG4=proteoglycan 4; AP=alkaline phosphatase; FSTL1=follistatin-like 1; CEACAM1=carcinoembryonic antigen-related cell adhesion molecule 1; ANPEP=alanyl (membrane) aminopeptidase; CRP=C-reactive protein; Hb=hemoglobin; CADM1=cell adhesion molecule 1; POSTN=periostin, osteoblast specific factor; CP=ceruloplasmin; AZGP1=alpha-2-glycoprotein 1, zinc-binding. 3 Fig. 2 A five-signature predictor for survival of castration-resistant prostate cancer (CRPC) patients. (A) The data matrix obtained by the analysis of 87 variables in 57 patients was subjected to random survival forest to obtain information on the predictive importance of each single variable. A representative bar chart shows the 87 variables in decreasing order of importance. (B–F) Graphs from single biomarkers belonging to the best signature for predicting survival in CRPC patients. CRP=C-reactive protein; AP=alkaline phosphatase; THBS1=thrombospondin 1; MME=membrane metallo-endopeptidase; PVRL1=poliovirus receptor-related 1; EFNA5=ephrin-A5; PSA DT=prostate-specific antigen doubling time; Hb=hemoglobin. [1] nomogram (blue) and the newly identified signature (red) in our dataset. European Urology  , DOI: ( /j.eururo ) Copyright © 2011 European Association of Urology Terms and Conditions
