2 Figure 1 Endogenous C57BL/6 (left) and MRL/MpJ (right) MSCs (Sca1+/CD140a+) after cartilage injury (Scale bar = 200μm).In contrast, MRL/MpJ mice demonstrated positive staining for MSCs within the defect and adjacent to the synovium as early as 1 hour after injury. Furthermore, at 2 weeks after injury, Sca-1+ cells were observed in the defect with a decreasing presence in and around the defect at 4 weeks after injury (Figure 1, D-F). To determine if increased numbers of MSCs were present before injury in MRL/MpJ mice, uninjured joints were examined. To control for differences in number of endogenous MSCs in both strains immediately after injury, and to determine if synovial MSCs play a role in cartilage repair, 10,000 GFP+ MSCs from either a C57BL/6 or MRL/MpJ mouse were injected into a C57BL/6 after joint injury. When C57BL/6 GFP+ MSCs were injected, little/no GFP staining was observed in the defect over the 4 week period (Figure 3, A-B).
