Figure 1 Endogenous C57BL/6 (left) and MRL/MpJ (right) MSCs (Sca1+/CD140a+) after cartilage injury (Scale bar = 200μm).In contrast, MRL/MpJ mice demonstrated positive staining for MSCs within the defect and adjacent to the synovium as early as 1 hour after injury. Furthermore, at 2 weeks after injury, Sca-1+ cells were observed in the defect with a decreasing presence in and around the defect at 4 weeks after injury (Figure 1, D-F). To determine if increased numbers of MSCs were present before injury in MRL/MpJ mice, uninjured joints were examined. No differences were observed in MSC staining in MRL/MpJ mice compared to C57BL/6 mice (Figure 2, A-B). Osteoarthritis and Cartilage , S13-S14DOI: ( /j.joca ) Copyright © Terms and Conditions

Figure 2 Endogenous C57BL/6 (upper) and MRL/MpJ (lower) MSCs (Sca1+/CD140a+) in uninjured cartilage (Scale bar = 200μm). To control for differences in number of endogenous MSCs in both strains immediately after injury, and to determine if synovial MSCs play a role in cartilage repair, 10,000 GFP+ MSCs from either a C57BL/6 or MRL/MpJ mouse were injected into a C57BL/6 after joint injury. When C57BL/6 GFP+ MSCs were injected, little/no GFP staining was observed in the defect over the 4 week period (Figure 3, A-B). Figure 3 Exogenous MSCs from a C567BL/6 (left) and MRL/MpJ (right) mouse injected into C57BL/6 mice (Scale bar = 200μm). However, when MRL/MpJ GFP+ MSCs were injected, GFP staining was found within the defect at 2 weeks, but was absent at 4 weeks (Figure 3, C-E).
