Analyzing plant extracts
4
Extract A
5
Extract C
7
Most of her extract A spilled into extract C and now she had only a little bit of extract A, extract B and a mixture of extracts A and C.
9
How would she analyze the pigments present in the respective extracts?
10
Hypothesis If extract A is analyzed and determined then the A+C mixture can be analyzed and extract C can be determined
11
Controls Positive Control Negative control:
Wavelengths between 401nm and 650nm Negative control: Wavelengths between 0nm and 400 nm Wavelengths above 650nm
12
Materials A, B, and A+C sample Chromatography paper Capillary tubes
Beakers Separation solvent Scissors Spectrophotometer and cuvette
13
Methods Plant extracts A, B, and A+C were placed on chromatography paper by use of capillary tubes 15 mL of separation solvent was put in beaker The chromatography paper was rolled into a cylinder and placed into the beaker with the solvent The chromatography paper was allowed to sit in the beaker for 15 minutes, then air dried
14
Methods (continued) The chromatography paper was divided into separate bands and each section cut into small pieces Each section’s pieces were put into designated test tubes with 3mL of isopropyl alcohol and allowed to sit The test tube solutions were tested for light absorption using a spectrophotometer at wavelengths from 400nm to 720 nm

Hypothesis was accepted
Results Hypothesis was accepted Chlorophyll A absorbs more light between range. The highest OD level was at a 440 wavelength for Chlorophyll A. Chlorophyll B absorbs more light between range. The highest OD level was at a 480 wavelength for Chlorophyll B. Chlorophyll C absorbs more light between range. The highest OD level was at a 580 wavelength For Chlorophyll C.
18
Conclusion The longer the wavelength the lower the energy and the farther it moves away from the pencil line.
