Figure 1 Label-free multiphoton microscopy of a fresh biopsy of human control colon tissue. The images represent thin optical sections recorded (A) in the epithelial layer (depth: 30 μm), (B) at the basal lamina (depth: 40 μm) and (C) in the lamina propria (depth: 50 μm). Detected signals originate from autofluorescent proteins like NADH (shown in green color) and flavins (red) and from second harmonic generation in collagen fibers (blue). Due to their specific spectral signatures, these signals can be recorded in individual color channels that allow to distinguish between epithelial cells (green) and immune cells (red) and to assess the amount of extracellular matrix (blue).
