9
BONE MARROW : The Site of Leucopoiesis
Vascular compartment Extensive network of sinusoids . Hemopoietic compartment Islands of hematopoietic cells (Hemopoietic cells in various stage of maturation )
11
HEMOPOIETIC GROWTH FACTORS Table 10-6 Gartner colour text histology page 242
Regulated the hemopoiesis Produce by spesific cells Acts on spesific stem cells, progenitor cells, and precursor cells The route to deliver growth factor : Via blood stream Secrete near the hemopoietic cells Direct cell-cell contact Induced rapid mitosis or and differentiation Most ot them are glycoproteins
12
GRANULOCYTOPOIESIS
13
GRANULOCYTOPOIESIS Formation of the granulocytes (neuthrophil,
eosinophil, and basophil) All of Granulocyte progenitor cell derive from CFC-S Granulocyte Progenitor : CFU-Eo : eosinophil lineage CFU-Ba : Basophil lineage CFU-GM CFU-G : Neutrophil line CFU-M : monocyte line
15
Maturation of Granulocyte
Granulocyte maturation divide into six stages Structural Changes : A decreased in cell size Condensation of nuclear chromatin Changes in nuclear shape An accumulation of cytoplasmic granule
16
Maturation of Granulocyte (Table 10-8 Gartner colour text histology page 242)
17
GRANULOCYTOPOIESIS Mature granulocyte leave the bone marrow then enter the circulation→ connective tissue
18
Monocytopoiesis
20
MONOBLAST Large cell (15-25 ) Nucleus oval
Cytoplasm without granule or few azurophlic granule Differentiate to the Promonocyte
21
PROMONOCYTE 16-18 µm in diameter Kidney shape nucleus
Cytoplasm contain azurophilic granule Divide twice in the course of their development into monocytes
22
MONOCYTE ON BONE MARROW
Mature monocytes enter the bloodstream → circulate. Monocyte enter the connective tissues, where they mature into macrophages.
23
LYMPHOPOIESIS
24
LYMPHOPOIESIS CFU-Ly divides form the CFU-LyB and CFU-LyT
CFU-LyB migrate to “bursa-equivalent location”, divided → B lymphocytes CFU-LyT undergo mitosis migrate to the Thymus → T lymphocytes

CONCLUSION Leucocytosis is formation of WBC, occurs in Bone Marrow
Granulocyte and monocyte derive from Myeloid stem cells, whereas limfocyte from Lymphoid Stem cell Leucocytosis involved the series of mitosis and differentiation Leucocytosis are regulated by several growth factor and Interleukin
