Top: Neutrophil expansion. When highly active, tumor-specific CD8+ T cells seed the tumor and start recognizing tumor antigens, release of HGF induce GMP expansion and neutrophil generation in the bone marrow, followed by release of c-MET+ neutrophils in the blood. These two events are blocked by c-MET inhibition. Bottom: Neutrophil commitment. The local release of factors from activated T cells (i.e., IFN-γ) turns neutrophils into enemies able to suppress the very same lymphocytes, in part through NO production and PD-L1 expression, within the tumor environment and draining lymph nodes. c-MET inhibition indirectly interferes with these processes by limiting the neutrophil arrival.
