Most of the Spo11 accessible sites in S. cerevisiae are nucleosome depleted regions (NDR) upstream of transcription start sites (TSSs). The Set1 complex interacting with RNA Pol II modifies the +1 nucleosome and promotes H3K4me3 (green star). The Spp1 protein, which is localized to the chromosome axis through an interaction with Mer2, is able to bind H3K4me3 (arrow). This promotes the tethering of the chromatin loop to the axis and Spo11 interaction with the Rec114/Mei4/Mer2 complex.
