Substance P neurons in the dorsal root ganglia (DRG) send peripheral projections to the gut wall, where they innervate blood vessels and other structures, and to the spinal cord, where they interact in laminae II and III with neurons that transmit pain sensations to the brain. The present knockout experiments indicate that the most important spinal receptors mediating pain and the most important vascular and pancreatic receptors mediating neurogenic inflammation and pancreatitis are neurokinin 1 (NK1) receptors.
