Figure 1 Differential protein-protein interaction between ADPRT and XRCC1 variants in human cancer cells. Myc-XRCC1-399Arg (designated as R) or Myc-XRCC1-399Gln (designated as Q) was respectively coexpressed with either Flag-ADPRT-762Val (designated as V) or Flag-ADPRT-762Ala (designated as A) in human breast cancer cell line MCF-7 cells (designed as RV, QV, RA, and QA, respectively) (A), gastric cancer cell line MGC-803 cells (B), and esophageal adenocarcinoma cell line SEG-1 cells (C) for 24 hours. Similar expression levels of different XRCC1 and ADPRT variants were detected in cell lysates by immunoblotting with anti-Myc, anti-Flag, and anti-β-actin antibodies (I). The cell lysates were preimmunoprecipitated with mouse control IgG followed by immunoprecipitation with anti-Myc monoclonal antibody (II) and with anti-Flag M2 monoclonal antibody (III). The immunoprecipitates including IP-IgG and IP-Myc or IP-Flag (lanes 1–8) as well as the RV lysate (lane 9) were subjected to 10% SDS-PAGE and immunoblotting analysis with Myc or Flag antibody.
