2 Figure 1 Epitope spreading. Tumor cell death (1) releases antigens which are phagocytosed by local dendritic cells (DCs) (2). DC activation is enhanced by IFNg and by CD40:CD40L interactions with CARTmeso cells. Activated DCs travel to the draining lymph node (3) where they engage naïve tumor-specific CD8 and CD4 T cells. Activated CD4 T cells may express CD40L and provide help for B cell class switching and antibody production (4). Tumor-specific effector CD4 and CD8 T cells and antibodies are recruited to the tumor microenvironment for perpetuation of a polyfunctional immune response (5).
