Figure 3 Expression levels of miR-1 and miR-133 in the stomachs of mice with H pylori infection. (A) Expression levels of miR-1, miR-133a, and miR-133b were analyzed by quantitative reverse transcriptase polymerase chain reaction using the TaqMan microRNA assay in the stomachs of control (blank bar; n = 7) and H pylori–infected mice (filled bar; n = 7). Expression levels were normalized against U6 RNA expression. All reactions were done in duplicate and expressed as mean values ± standard deviations. *P < .005 compared with controls. (B) Expression levels of miR-1, miR-133a, and miR-133b in clinical samples from the gastric antrum of patients with or without H pylori infection. Expression levels of miR-1, miR-133a, and miR-133b were significantly decreased in patients positive for H pylori infection (n = 14), relative to patients who were negative (n = 8; miR-1, P < .005; miR-133a, P < .05; miR-133b, P < .01). Figure 4 Characterization of hepatic stem cell marker expressions in Hep3B cells. The majority of ATP-dependent binding cassette (ABC)B5+ cells are also GEP+, and these GEP+ABCB5+ double positive expressed CD133 and EpCAM, both stem cell markers. Suppression of ABCB5 by the siRNA approach effectively decreased ABCB5 expression, reduced the population of cells co-expressing GEP, and diminished the cell population expressing the hepatic stem cell markers CD133 and EpCAM.
