Retroviral vector propagation in helper cells and structure of a prototypical vector. A, propagation of retroviral vectors in helper cells. Helper cells produce the viral proteins (shown as black ellipses and white circles) that are used to assemble viral particles containing RNA transcribed from the viral vector. Target cells do not express viral proteins and cannot generate viral particles containing the vector RNA. B, cis-acting elements needed in a prototypical retroviral vector.
