In the Lgr5+ intestinal stem cell (ISC), Wnt/Rspo signals induce Ascl2, which undergoes positive auto-regulation and conversion from an Ascl2 “off” state to an Ascl2 “on” state, consistent with a bistable switch. Wnt/Rspo regulation of Ascl2 is also governed by hysteresis where previous Wnt/Rspo exposure enhances the ability of Wnt/Rspo to induce the Ascl2 “on” state, thereby stabilizing the stem cell identity of the Lgr5+ ISC. Ascl2 further cooperates with β-catenin/Tcf4 to transcriptionally activate stem cell loci that define Lgr5+ ISC identity. Cell Stem Cell , DOI: ( /j.stem ) Copyright © 2015 Elsevier Inc.
