Figure 1 Lamin A Promotes Longevity by Regulating the Function of Two Key Longevity Assurance Factors: NRF2 and SIRT6 In the wild-type cells, lamin A positively regulates SIRT6 activity. Lamin A may also promote the formation of active NRF2-SIRT6 complexes, which in turn ensure efficient activation of oxidative stress response pathways, DNA repair, and heterochromatin maintenance. In the cells expressing progerin, NRF2 and SIRT6 are sequestered in nonfunctional state at the nuclear membrane leading to impaired oxidative stress response, genomic instability, and loss of heterochromatin.
