Fig. 1 Differential gene expression in RCC. (A) The density of 18s and 28s RNA was nearly equivalent in lab-on-a-chip. (B) The upper image represents internal control spots. The bottom picture represents gene profiling overlaid images of 0.12K cDNA slide hybridized with the conventional RCC (Cy3; green) and chromophobe RCC (Cy5; red). Arrow (yellow) and arrowhead (white) indicate intense green signals, which point at MMP9, MMP2, respectively. (C) Each mean value represents normalized intensity of the Cy3/Cy5 ratio. Notice a high level of MMP9 in the conventional RCC. European Urology , DOI: ( /S (03) )

Fig. 2 Differences in the mRNA expressions of MMP2 and MMP9 in RCC. The MMP9 transcript level in RCC is variable at the constant expression of β-globin as an internal control, in formalin-fixed paraffin-embedded RCCs. Note the constant expression of MMP2 mRNA. Almost all cases showing evidence of MMP9 mRNA expression were of ‘conventional’ RCCs. European Urology , DOI: ( /S (03) )

Fig. 3 Comparison of mRNA expression with the gelatinolytic level of MMP9. (A) In the RT-PCR of MMP9, cases 2 and 5 exhibited apparent bands at 262bp. (B) Pairs of renal tumor tissue (T) with the corresponding normal tissue (N) were analyzed by gelatin zymography. MMP9 gelatinolytic activity was apparent in ‘conventional’ and ‘papillary’ RCCs (lanes 2, 3, 4, and 5), which was more evident by image analysis. The case shown in lane 1 was of chromophobe RCC of stage III (renal vein thrombi); lane 2, ‘conventional’ RCC of stage III (renal vein thrombi); lane 3, ‘conventional’ RCC of stage I; lane 4, ‘papillary’ RCC of stage I; lane 5, ‘conventional’ RCC of stage II; lane 6, ‘oncocytoma’; lanes 7, and 8, ‘chromophobe’ RCC of stage I. Notice a strong expression of MMP9 mRNA in advanced stage of the ‘conventional’ RCC (lanes 2 and 5). European Urology , DOI: ( /S (03) )

Fig. 4 Kaplan-Meier survival curve according to MMP9 transcript expression. Patients with high level of MMP9 transcript showed significantly shorter survivals (p= by log-rank test). A black circle means negative transcript; open circle, weak signal; triangle, strong signal for MMP9 mRNA (time scale units represent a month). European Urology , DOI: ( /S (03) )
