Mutations affecting HOIP–UBA and HOIL‐1L–UBL interaction that compromise NF‐κB activation. (A) NF‐κB activation by HOIP–UBA and HOIL‐1L–UBL interaction. Expression of HOIL‐1L and HOIP mutants (right panel) and luciferase activity of 293T cells transfected with the indicated plasmids and the 5 × NF‐κB reporter (left panel) were assessed. Values of luciferase activity are mean±s.e.m., n=3. Statistical analyses were performed using a two‐tailed unpaired Student's test, *P<0.01. (B) HOIP–UBA and HOIL‐1L–UBL formed complex through contact 1‐mode of interaction. Lysates and immunoprecipitates with anti‐HA and anti‐Myc antibodies from cells that expressed the indicated plasmids were probed as indicated.
