1 Rewiring Mitochondrial Pyruvate Metabolism: Switching Off the Light in Cancer Cells?

2 Figure 1 MPC Expression and Metabolic Targeting of Mitochondrial Pyruvate High MPC expression (green) is associated with more favorable tumor prognosis, increased pyruvate oxidation, and reduced lactate and ROS, whereas low expression or mutated MPC is linked to poor tumor prognosis and increased anaplerotic generation of OAA. Dual targeting of MPC and GDH with small molecule inhibitors may ameliorate tumorigenesis in certain cancer types. Abbreviations: MPC, mitochondrial pyruvate transporter; GDH, glutamate dehydrogenase; ROS, reactive oxygen species; TCA, tricarboxylic acid (cycle); OAA, oxaloacetate; PC, pyruvate carboxylase; UK5099, 2-Cyano-3-(1-phenyl-1H-indol-3-yl)-2-propenoic acid; CHC, α-Cyano-4-hydroxycinnamate; EGCG, epigallocatechin gallate.
