Fig. 1 iHAst-specific proliferative responses in PBMCs from 29 healthy adult subjects. PPD from M. tuberculosis was used as a positive control antigen and noninfected Caco-2 cells as a negative control antigen. Proliferative responses were measured by [3H]thymidine uptake for the last 20 hours of a 7-day antigenic stimulation. Rate of proliferation is given as SI (Antigen-Specific Proliferation of PBMCs ÷ PBMC Proliferation Without Antigen Stimulation). Horizontal lines indicate mean value of SI for each antigen. Fig. 2 iHAst-specific proliferative responses and HLA restriction of small intestinal TCLs from 4 adult subjects. Responses were measured by [3H]thymidine uptake hours after iHAst stimulation. Autologous PBMCs were used as APCs. Inhibition of iHAst-specific responses was performed by the addition of anti-HLA MAbs (10 mg/mL) 30 minutes before addition of the TCL. Data for each T-cell line are from one representative experiment (out of 3). Gastroenterology , DOI: ( /S (98) ) Copyright © 1998 American Gastroenterological Association Terms and Conditions

Fig. 3 HLA restriction pattern of 6 iHAst-specific CD4+ TCCs derived from duodenal biopsy specimens from 2 adult subjects (As3, HLA-DR2,DQ6/DR4,DQ7; and As5, HLA-DR2,DQ6/DR8,DQ4). As APCs, autologous PBMCs and allogeneic HLA-typed PBMCs were used. Responses were measured by [3H]thymidine uptake hours after iHAst stimulation. Inhibition of iHAst-specific responses was performed by addition of anti-HLA MAbs (10 mg/mL) 30 minutes before addition of TCC. Data given on each TCC are from one representative experiment (out of 2). Gastroenterology , DOI: ( /S (98) ) Copyright © 1998 American Gastroenterological Association Terms and Conditions
