Arsenic and retinoic acid bind the promyelocytic leukaemia (PML) and retinoic acid receptor-α (RARα) moieties of the PML–RARα fusion oncoprotein, respectively, triggering its degradation. This allows PML to activate p53 specifically in acute promyelocytic leukemia (APL) blasts, resulting in leukaemia eradication.
