Introduction-DNA The genomes of almost all organisms are DNA, the only exceptions being some viruses that have RNA genomes. Genomic DNA molecules constitutes the total genetic information of an organism. They are generally large, and in most organisms are organized into DNA–protein complexes called chromosomes.
5
DNA Extraction Studying DNA is very important, which aids in disease prevention, diagnosis and treatment. The first step to study DNA is to do extraction !! If you want to study the genetics of Alzheimer disease for example, From where do you think DNA should be extracted?
6
DNA Extraction Many different methods and technologies are available for the isolation of genomic DNA In Blood DNA is found in ……….cells
7
DNA Extraction Methods
1.Salting-out 2.Organic extraction 3.Binding of the DNA to a solid-phase support (either anion-exchange or silica technology).

In Our Experment-QIAamp DNA Blood Mini Kits (silica technology)
The QIAamp DNA purification procedure comprises 4 steps and is carried out using QIAamp Mini spin columns in a standard microcentrifuge which contain a seleca membrane that bind to DNA
9
Principle: No phenol-chloroform extraction is required.
DNA binds specifically to the QIAamp silica-gel membrane while contaminants pass through. PCR inhibitors, such as divalent cations and proteins, are completely removed in two efficient wash steps, leaving pure nucleic acid to be eluted in either water or a buffer provided with the kit.
