Figure 1 Structure of the At4g14850 Gene and Encoded PPR Protein.
(A) Schematic representation of gene structure from ATG to stop codon. Exons are indicated by white rectangles and the intron by a hatched line. T-DNA insertions are indicated by black triangles and nucleotide polymorphisms in has2/mef11-6 mutant sequence are shown in bold. (B) Schematic structure of the MEF11 protein, which includes P, L, S motifs, an E-like domain, and a DYW domain (adapted from Verbitskiy et al., 2010b). Mutation positions are shown. 3 Figure 2 Leaf Temperature of mef11 and pp2c Mutants Visualized by Thermal Imaging. False color infrared images of the temperature of drought-stressed plants are shown. Leaf temperature of (A) mef11 mutant alleles compared with wild-type Col-0 (WT), (B) multiple pp2c mutants compared with wild-type, (C) mef11-6 compared with pp2c mutants. Watering had been withheld for 3 days before infrared imaging. Scales indicate temperature (°C). Molecular Plant 2015 8, DOI: ( /j.molp ) Copyright © 2015 The Author Terms and Conditions

Figure 3 Germination Rates of mef11 Mutant Seeds Compared with the Wild-Type (WT) and Quadruple Qpp2c Mutant (hai1-1 pp2ca-1 hab1-1 abi1-2). (A) Germination of freshly harvested seeds was scored during imbibition at 25°C under continuous light on solidified water (0.5% [w/v] agarose). (B) Germination of after-ripened seeds was scored in the same conditions after a 3-day stratification (incubation at 4°C in the dark). Germination was based on testa rupture; averages and SE (n = 3) are shown. Similar results were obtained using seeds from at least two independent cultures. 5 Figure 4 Inhibition of mef11 Mutant Germination by ABA, Paclobutrazol, NaCl, and Mannitol, Compared with Wild-Type (WT) and Quadruple Qpp2c Mutant (hai1-1 pp2ca-1 hab1-1 abi1-2). Germination of stratified seeds on solidified water (0.5% [w/v] agarose), containing 0.2 μM ABA (A), 1 μM paclobutrazol (B), 100 mM NaCl (C), or 300 mM mannitol (D), was scored over 5 days after transfer to 25°C in the light. Results were obtained using three biological replicates for each genotype. Average and SE of seedlings with green cotyledons (A and B) and seeds exhibiting testa rupture (C and D) are shown, relative to maximal germination in the absence of inhibitor. Similar results were obtained using seeds from at least two independent cultures. Molecular Plant 2015 8, DOI: ( /j.molp ) Copyright © 2015 The Author Terms and Conditions

Figure 5 RNA Editing at nad4, ccmFN2, and cox3 Sites in Wild-Type and mef11 Mutant Alleles. Sequence chromatograms of edited codons obtained after RT-PCR of cDNA fragments. The amino acid residues are indicated above. Molecular Plant 2015 8, DOI: ( /j.molp ) Copyright © 2015 The Author Terms and Conditions

Figure 6 Analysis of the Relative Abundance of Respiratory Complexes in mef11 Mutants and Wild-Type. (A) Crude membrane extracts from wild-type and mef11 mutants separated on BN-PAGE gel and stained with Coomassie blue. (B) NADH dehydrogenase in-gel staining revealing complex I activity. (C) Immunoblot using antibodies to RISP revealing complex III. (D) Immunoblot using antibodies to COX2 revealing complex IV. The different respiratory complexes are identified by their roman numeral. 8 Figure 7 OH° Content of mef11 Mutant Plants Compared with Wild-Type (WT) and the Quadruple Qpp2c Mutant (hai1-1 pp2ca-1 hab1-1 abi1-2). Data are averages of three biological replicates with SE. No significant differences with wild-type were detected using the non-parametric Mann–Whitney rank sum test. Similar results were obtained with plants from two independent cultures. 9 Figure 8 H2O2 (A) and NO (B) Content of mef11 Mutant Imbibed Seeds Compared with Wild-Type (WT) and the Quadruple Qpp2c Mutant (hai1-1 pp2ca-1 hab1-1 abi1-2). Data were obtained using three biological replicates; averages and SE are shown. Asterisks indicate significant differences from wild-type using the non-parametric Mann–Whitney rank sum test (P value = 0.05). DSW, dry seed weight before imbibition.
