SUPPLEMENTAL FIGURE 2. TNFa is neither interfering with cell surface receptor ligation, nor with detection antibody binding to porcine aortic endothelial cells (PAEC)-KO. PAEC-KO were cultured with 100 U/ml human TNFa, in the presence of heparin (20 mg/ml) during culture and/or antibody incubation. TNFa stimulation resulted in increased expression of CD62E (A) and CD106 (B) on PAEC-KO (solid histogram) that was unaffected by both heparin preincubation (open dashed line) or coincubation (open dotted histogram), and by incubating the cells with heparin immediately before the addition of detection antibodies (open dashed-dotted histogram).
