Figure 10 | The energy depletion hypothesis. Mutations implicated in hypertrophic cardiomyopathy are purported to increase the energy cost of force production by the sarcomere. Consequently, less energy is available for other critical functions of the cardiac myocyte, such as Ca2+ reuptake into the sarcoplasmic reticulum. Chronic energy depletion leads to myocyte dysfunction, elevation of cytosolic Ca2+, and 5′-AMP-activated protein kinase (AMPK) activation, with downstream hypertrophy.
