The relative sensitivity of mitochondrial probe content to 20 mV decreases in ΔψM and ΔψP was modeled for TMRM (solid line) and second-order MitoClick accumulation (dashed line) as a function of incubation time using Equation 18 in Gerencser et al. (2012). The dotted trace predicts the effect of an alternative 1 min pulse labeling with MitoClick precursors followed by complete wash off and later tissue harvesting at different times. Modeling assumed that 10% of the cell is mitochondrial matrix, the probe is at 90% equilibrium with ΔψP at 60 min, Δψp = −60 mV, and ΔψM = −140 mV.
