1 Differential expression of elastin assembly genes in patients with Stanford Type A aortic dissection using microarray analysis  Bernice L.Y. Cheuk, PhD, Stephen W.K. Cheng, MSc, FRCS, FACS  Journal of Vascular Surgery  Volume 53, Issue 4, Pages e2 (April 2011) DOI: /j.jvs Copyright © 2011 Society for Vascular Surgery Terms and Conditions

2 Fig 1 Elastic fiber fragmentation in control (A) and dissected aortas (B, C, and D), was shown by Movat pentochrome staining (under 200× magnification). Using this staining method, elastic fibers (blue-black color), nuclei (blue), collagen (red), and muscle (yellow) were differentially stained. The black bar indicates 5μm. Destructive architecture within the meidal layers with lower elastin content was obvious in dissected aortas. The elastic fiber fragmentation was largely interlamellar and created “cystic spaces” as evaluated by this staining method. 3 Fig 2 Validation of microarray data by semiquantitative reverse transcriptase polymerase chain reaction (RT-PCR). The expression patterns of fibulin-1, matrix metalloproteinase-9 (MMP-9), and elastin were consistent with the expression pattern shown in microarray experiments. Here, β-actin was used as housekeeping gene, which was assessed for the relative abundance in both control (control 1-4) and aortic dissection tissues (AD1-4). 4 Fig 3 Analysis of protein expression patterns of fibulin-1, matrix metalloproteinase-9 (MMP-9), and elastin at protein levels. Representative western blot results of control, aortic dissection, and nondissected tissues was shown. Here, β-actin was used as housekeeping protein, which was assessed for the relative protein abundance in the three groups of tissues. 5 Fig 4 Densitometric analysis of gene (A) and protein (B) expression levels of fibulin-1, matrix metalloproteinase-9 (MMP-9), and elastin was conducted. Relative expression levels of interested genes and proteins were calculated by dividing the expression level of target gents with the expression level of β-actin of the same RNA or protein sample. Data were personated as the mean ± standard deviation. AD, Aortic dissection. (*P < .001, was the statistical significant value between dissected aortas and control, and **P < .001 was the statistical significant value between nondissected aortas and control. No statistical difference was found between the expression level of dissected aortas and nondissected aortas.)
