Cells are crosslinked with formaldehyde in order to introduce reversible covalent bonds between proteins and DNA. After cell lysis and shearing of DNA, the protein of interest is immunoprecipitated together with bound DNA fragments, which are then decrosslinked, purified, labeled, and hybridized to microarrays. Finally, after scanning, ChIP-on-chip signals are plotted against genome coordinates to visualize the protein binding sites.
