(A) mtMSI in the (C)n tract was detected by PCR-SSCP analysis. Cases 17, 23, and 28 show differing mobilities in carcinoma tissues (C) compared with normal counterparts (N). These alterations were detectable in both single-strand (ss) and double-strand (ds) DNA patterns. Case 28 was defined as mtMSI negative because the mutant sequence could not be determined by direct sequencing. (B) Mutations in the mitochondrial tRNA (Leu) and ATPase 6 genes were detected by PCR-SSCP analysis. Cases 5 and 23 showed mobility-shift bands (arrowheads) in comparison with normal counterparts, whereas others (4 and 24) did not show any band shifts.
