Figure 1 A Simplified View of eSPAN
eSPAN is a sequential ChIP protocol. Synchronized yeast are released into S phase with a pulse of bromodeoxyuridine (BrdU) to label the nascent DNA. An antibody for the protein of interest is used for the first IP step, while anti-BrdU is used in the second IP step. The 3′-end of each strand is then ligated to an oligonucleotide so that the source strand information would be retained and sequencing reads can be mapped to the yeast reference genome and any biases toward the leading or lagging strand are calculated.
