Figure 1 Model of Lymph-Node-Resident and Peripheral Tfh Cells
In this model, Tfh cells classically reside in the germinal center of the lymph node and express PD-1, CXCR5, and ICOS on the cell surface and the transcription factor Bcl-6. Peripheral Tfh (pTfh) cells can be found in the blood and express PD-1 and CXCR5 on the cell surface and, in this issue of Immunity, Schultz et al. (2016) report that they can be defined by IL-21 production ex vivo. Tfh and pTfh cells are transcriptionally and functionally related, playing a critical role in antibody class-switching, somatic hypermutation, and affinity maturation.
