In IDH-wild-type glioma (top panel), the FIP1L1 enhancer activates FIP1L1 expression. PDGFRA expression is insulated from this effect (via CTCF insulator binding). In contrast, IDH mutant tumors (lower panel) show aberrant methylation, reducing the binding of the insulator and resulting in elevated expression of PDGFRA to promote gliomagenesis.
