Figure 1 A Schematic Model for the Conversion of Ly6C+ into Ly6C− Monocytes in the Mouse Monocytes receive signals from the bone marrow niche that instruct the activation of the gene expression program characteristic of Ly6C+ monocytes. Once in the blood, Ly6C+ monocytes can extravasate in response to homeostatic or danger signals, enter tissues, and terminally differentiate into macrophages. After the Ly6C+ program is extinguished, a distinct program under the control of the transcription factors C/EBPβ, KLF2, and NR4A1 is derepressed: the resulting Ly6C− monocytes remain in the blood to exert endothelium patrolling activities.
