This diagram shows the mechanism proposed by Zambetti et al. to illustrate the contribution of Sbds mutation in mesenchymal stem/progenitor cells (MSPCs) to reduced bone formation and impaired hematopoiesis (myelodysplasia) in Shwachman-Diamond Syndrome. The loss of Sbds in osterix+ MSPCs (right panel) impairs osteoblastic differentiation and induces p53. S100A8/A9 secreted by MSPCs activates TLR4 in hematopoietic stem/progenitor cells (HSPCs) and causes mitochondrial hyperpolarization and an increase of ROS and DNA damage response.
