Figure 1 Microbiome analysis and identification of the Propionibacterium genus by two different 16S rRNA marker gene primer sets located in the V3–V6 region. One healthy individual was sampled three times at the upper buttock over a period of 3 years. Two different primer sets were used: (a) the 338F-1061R primer set (for sequences of this primer set, see Zeeuwen et al., 2012) and sequenced on a 454 platform and (b) the primer pair combination 357F (5′-TCGTCGGCAGCGTCAGATGTGTATAAGAGACAGCCTACGGGAGGCAGCAG-3′) and 802RV2 (5′-GTCTCGTGGGCTCGGAGATGTGTATAAGAGACAGTACNVGGGTATCTAAKCC-3′), in which the italicized sequence is the Flag sequence and the bold sequence is the 16S bacterial primer sequence, which was sequenced on a MiSeq Illumina platform (BaseClear BV, Leiden, The Netherlands). rRNA, ribosomal RNA; V, hypervariable region of the 16S rRNA gene.
