1. Today’s Goals Learn techniques to transfer nematodes by “chunking”
Learn to make a worm “pick”
For moving individual worms
Learn to Pick worms and transfer them from plate to plate

3. Chunking:
Observe C. elegans under dissecting microscope to make sure they are alive
Sterilize a scalpel
Flame it. No Ethanol.
Cut a chunk of agar and transfer to a fresh, OP50 seeded plate
Make 2 plates per person
Put inverted in incubator in back of class that is NOT on
We will check on the results next class.

4. Picking Worms Demo – Make a worm pick www.silencinggenomes.org
Make 1 per person, Instructor will cut the wire for you.

5. Practice Picking Worms

6. Safety Concerns: Wear glasses and gloves!
Use great care when flaming worm picks
Only need to flame for a MOMENT
DON’T hold it too long or you will melt the pipet!!!
Be sure to flame pick when finished - or bacterial contamination will result
