Optical density of binding measures 0.37 ± C, Nonspecific binding for insulin. Excess of insulin was used to compete with 125I-IGF-I for its binding sites. Optical density measures 0.15 ± D, Nonspecific binding for IGF-I. Excess of unlabeled IGF-I was used to compete with 125I-IGF-I for binding sites. Optical density measures 0.05 ± 0.007, which is not different from optical density of film measured at 0.04 ± Note difference in nonspecific binding between C and D. Difference in optical density of C and D autoradiographs is statistically significant with p < with paired t test.
