TRIF Triggers STING Activation and Type I Interferon Secretion
STING is activated by cyclic dinucleotides released from bacteria or synthesized by cGAS upon microbial infection. Wang et al. (2016) demonstrate that TRIF directly binds to STING, promoting its dimerization, intermembrane translocation, and signaling. TRIF deficiency enhances HSV1 viral replication and causes a severe pathology. Impaired STING signaling may be a possible explanation for the herpes simplex encephalitis associated with TRIF mutations in humans.
