Oncogenic β-catenin and MMP-7 (matrilysin) cosegregate in late-stage clinical colon cancer  Andrei V. Ougolkov, Kaname Yamashita, Masayoshi Mai, Toshinari Minamoto  Gastroenterology  Volume 122, Issue 1, Pages (January 2002) DOI: /gast Copyright © 2002 American Gastroenterological Association Terms and Conditions

2 Fig. 1 Representative expression patterns of (A-C) β-catenin and (D) MMP-7 in colon adenocarcinoma. (A) Membranous expression of β-catenin was represented by delineating the carcinoma cell membranes. (B) NAd of β-catenin found in a well-differentiated adenocarcinoma. (C) In a tumor showing NAinv of β-catenin, NA of this molecule was exclusively detected in carcinoma cells at the invasion front. The carcinoma cells in the parts other than the invasion front of this tumor showed membranous expression but no NA. (D) In an MMP-7–positive tumor, this enzyme was expressed in the cytoplasm of carcinoma cells in the invasion front of the tumor. (A-D) Original magnification 50×. 3 Fig. 2 Immunohistochemical staining of a pair of serial sections from the same tumor in a mirror image clearly demonstrated colocalization of (A and C) nuclear β-catenin and (B and D) cytoplasmic MMP-7 in the same carcinoma cells in the invasion front. (C and D) Same parts of A and B, respectively, at higher magnification. Original magnifications: (A and B) 50x; (C and D) 100×. 4 Fig. 3 Comparison of (A and C) DFS and (B and D) OAS of the patients with colon cancer according to the different expression patterns of β-catenin and MMP-7 in the primary tumors. The vertical axis indicates survival rate. Kaplan-Meier survival curves showed a statistically significant survival disadvantage in (A and B) colon carcinomas with the NAinv pattern of β-catenin and in (C and D) those with MMP-7–positive tumors, compared with those with other expression patterns of these molecules. The significant differences (P values) between the respective groups of patients were calculated by the log rank test. Hazard ratio and 95% confidence intervals for the comparisons were calculated by the Cox regression model (Table 2). The expression patterns of β-catenin were classified as follows: NA (−), membranous expression and loss of expression; NAd, diffuse nuclear accumulation; and NAinv, nuclear accumulation in the invasion front of the tumor. MMP-7 (−) and MMP-7 (+) indicate absence and presence, respectively, of MMP-7 expression in the invasion front of the tumor.
