Fig. 2 SDS-PAGE/silver staining of nCA and rCA. Natural and recombinant albumin were both analyzed by SDS-PAGE/silver staining. nCA demonstrated a major band of greater than 60 kd. Both high and low molecular weight (MW) contaminations were detected. Culture supernatant–containing rCA is shown in lane 3 . A major band at the same molecular weight as nCA was observed. A contaminating band just below rCA was also present in control culture supernatant (not shown). Molecular weight markers contain a 10-kd ladder. Fig. 3 Comparison of IgE reactivity with nCA and rCA by means of immunoblot. Thirteen sera were analyzed on immunoblot for IgE reactivity with nCA and rCA. Sera can roughly be divided in 3 different reactivity patterns with both versions of albumin (A , B , and C ). Only the first group (left 5 lanes) recognized nCA and rCA (A) . None of the discrepant sera recognized CA at all (B and C ). For reference purposes, mAb 4E9 reactivity is shown in the last lane.
